Isolation of compounds from Cannabis sativa and their anti-inflammatory effect

The object of this thesis is a traditional and also controversial plant Cannabis sativa. The theoretical part is focused on ubiquitous polyphenolic compounds – flavonoids, also present in the studied plant. All previously researched and confirmed effects on human body are described in detail. The experimental part of this thesis describes the isolation and identification of substances of the chloroform and hexane yields of extract obtained from this plant. Chromatographic methods were used for isolation – thin layer and column chromatography and semipreparative HPLC. Obtained substances were characterized using UV, IR spectrophotometry and NMR. Then the anti-inflammatory activity of obtained substances and also of the ethanolic extract and its yields was observed. The result of the experimental part was the isolation of one pure substance which was identified as the cannabidiolic acid. Using the THP 1 assay, we demonstrated mild anti-inflammatory effect of the non-polar yields and isolated CBDA.

The object of this thesis is a traditional and also controversial plant Cannabis sativa. The theoretical part is focused on botanical side of this plant and its presently used medical forms. All previously isolated substances and their observed biological activity are described in detail. The experimental part of this thesis describes the isolation and identification of substances of the chloroform extract fraction obtained from this plant. Chromatographic methods were used for isolation – analytical and semipreparative HPLC. Obtained substances were identified using UV and IR spectrophotometry. Then the biological effect of obtained substances was observed on chosen microorganisms.
Hemp as an alternative fuel

Bachelor thesis as literary research deals with possibilities of using hemp as an energy crop and its development and sustainability in the future. Production of hemp bio-fuels in Czech Republic is in the beginning but increasing interest in alternative energy sources presumes development in this branch. Producing bio-fuels made of hemp biomass brings many positive benefits in liquid, gas or solid states. Liquid biodiesel made of hemp presents alternative source of energy that can be used in common diesel engines. Hemp hurds briquettes are the most used hemp fuel in our country. Hemp briquettes has perfect properties in comparison to other sources. High heat of combustion, low purchase price and environmental friendliness gives motivation to create new growing branches. Second most common use is biogas fuel. Biogas is produced in usual biofermenter using hemp biomass often with admixture of slurry. Hemp biogas properties are also very satisfactory. In Czech Republic is huge potential in development and production of hemp biogas due to large number of biogas plants. Despite financially demanding processing of the hemp biomass the amount of biomass grow during one year makes this plant one of the most profitable alternative source for production of bio-fuels.
Utilization of Cannabis Sativa in Teaching Chemistry

The diploma thesis deals with Cannabis sativa. The aim is to focus on possible utilization of Cannabis sativa in teaching chemistry. The theoretical part describes the botany of cannabis, overviews phytocannabinoids, substances contained in cannabis, and it also includes wide use of cannabis in agriculture and industry. The practical part deals with the selection and arrangement of a possible curriculum about chromatography as the most affordable technique for analysis of cannabis at school, including the formulation of school experiment methodology using the thin-layer chromatography of Cannabis sativa extract to demonstrate the presence of phytocannabinoids. The practical part also includes the scheme of an educational project on cannabis and describes the latest techniques, as well as the methodology of work with them, which are suitable for analysis of phytocannabinoids.

Diploma thesis entitled Contemporary Cannabis growth technology influence on yield and quality in the teoretical part deals with the legislation of cannabis cultivation, agrotechnology of cultivation, harvesting and hemp growing in the Czech Republic, in the European Union and in the world. In the practical part deals with experiments on the influence of arbuscular-mycorrhizal fungi on the content of selected cannabinoids in cannabis plants of the Kompolti and KC Dora varietes in controlled greenhouse conditions at the Institute of Botany of the CAS, v. v. i. and subsequently in the field of Sasov biofarm with Futura, Fibrol, KC Dora and Finola varietes. The effect of inoculation on the increase cannabinoids content has not been demonstrated.

The theoretical part of diploma thesis briefly focuses on botanical characteristics of cannabis, legislation of cannabis cultivation in Czech republic, agrotechnics and harvest of hemp. The cultivation technologies used in production and harvest of cannabis for pharmaceutical purposes are described in more details. In the practical part was established an experiment with cannabis plants. This experiment was carried in laboratory at department of plant biology at Mendel university in Brno. The experiment was carried out in two variants of light spectrum (white LED lights emitting complete visible spectrum and combination of blue and red LED in ratio 1:1), which were dividen into three groups with different trim method. In plants were observed parameters of quantum yield of electron transport of photosystem II, chlorophyll fluorescence, yield of dried flowers and selected secondary metabolites. In each of lighting and trim variants have been recorded various results. As results shown, white LED lights emitting a full visible spectrum appear to be more effective than the blue and red spectrum in ratio of 1:1. Effect of trimming in this experiment was not statistically conclusive.
Opinion of Pupils on Consumption of Marijuana: Prevention and Health Risks

The aim of this thesis is to sum up the information connected with marijuana and cannabis topic in relation to their effects, use and misuse. At the beginning of my research three hypotheses have been determined . They have been based on the electronic questionnaires which have been answered by respondents in lower secondary schools. The results of my thesis are shown in its applied part. According to the questionnaire the first hypothesis has been confirmed. 17,68 % of survey participants have tried marijuana at least once in their lives. The second hypothesis has not been taken to be true, because although there is a prevention in primary schools, it doesn't provide pupils ' awareness of this problem completely. The third hypothesis has been proved false as well, because more than 50 % of the respondents said that they have already learnt about cannabis in Science. Možnosti využití zbytků po zpracování technického konopí Cannabis sativa ve výživě brojlerových kuřat

The aim of this thesis was to evaluate the effects of addition of technical hemp (Cannabis sativa L.) on performance parameters of broiler chickens. A total of 105 broilers Ross 308 were divided into three groups. The groups were identified: Cannabis 5, Cannabis 15 and Control. Group Cannabis 5 was fed with 5 % hempseed cake in feed composition. Group of Cannabis 15 was fed with 15 % hempseed cake in feed composition. Group of Control was not fed with hempseed cake. As a response criterion was chosen weight gain, feed conversion, lean meat, carcass yield and blood liver enzymes concentration. In this thesis, we focus on assessing the impact of hempseed cakes on weight gain, feed conversion and the carcass yield. The increments in body weight were significant statistical differences (P < 0.05). Broilers fed with 15 % hempseed cake in feed composition grew slowly and had worse feed conversion. Carcass yield was not affected (P > 0.05) by content of hempseed cake in feed composition. Measured concentration of enzymes in groups was not affected by contents of hempseed cake.
